12/2004:
Sequencing of Unknown Genomic Regions

Medigenomix has successfully established a method obtaining a nucleotide sequence adjacent to known sequences in genomic DNA. Using minimal amounts of genomic DNA, we are able to systematically sequence large nucleotide segments employing a PCR based technique, established by our scientists. Thus we circumvent a shot-gun approach and its necessity to sequence many recombinant clones.

If you are interested in determining the sequence of, e.g., a cosmid insert (or other integration vector) or the integration site of a transposon our strategy is a very good alternative, reasonably priced and fast.

07/2004:
Medigenomix Receives Award for the Establishment of Relevant SNPs According to Gene Diagnostic and Pharmacogenetic

Medigenomix GmbH, a subsidiary of the Eurofins Group, received an award of about EUR 1 mio. from the German Federal Ministry of Education and Research within the framework of the "BioChance Plus” competition. The ministry thereby supports application-oriented high-risk research and development projects of small and medium-sized biotechnology enterprises. This programme aims at accelerating technology transfer from basic research and the formulation of market utilisation strategies. The successful proposal is one of only a few of about 220 submitted project drafts that have been chosen due to its individualised medicine concept. The proposal includes the establishment of innovative, ready-for-market, all-purpose DNA-tests for individual gene diagnostics to predict the course of disorders and the individual response to drug therapies. In cooperation with the University Hospital Essen polymorphisms in genes (SNPs) coding for G-proteins will be screened and validated. As G-proteins play a pivotal role in signal transduction processes from receptors to intracellular components in all human tissues, variations in their protein structure or expression influence all drug actions and the natural courses of common disorders.

With this project, Medigenomix expands its efforts and research capacities in the field of Pharmacogenetics by a further research area. Hitherto, Medigenomix offered analytical and pharmacogenetic consulting service for clinical trials in the biopharmaceutical industry using the acquired exclusive licenses of polymorphisms in G-proteins. With the support of the BioChance Plus Programme, the company is now able to carry out its own research in the field of pharmacogenetics in cooperation with Prof. Siffert’s group at the University Hospital Essen and will thus strengthen its competence in G-protein-polymorphisms. The technological and scientific know-how of both partners will be bundled and the validated diagnostic tests will be submitted for international patent applications. Thereby, Medigenomix will make an important contribution to individualised medicine.

Medigenomix is a renowned service laboratory for DNA analyses with longstanding experience in genotyping for a broad range of applications from breeding to forensics and medicine.

06/2004:
Expression Consortium Martinsried – Services from Genes to Proteins

ProteingelThe specialists for Genomics and Proteomics, Medigenomix GmbH and Toplab GmbH, Martinsried, recently founded the Expression Consortium Martinsried.

Within this consortium, a complete gene expression service from gene to protein is available for customers. The service encompasses all steps, starting with the design of an in silico DNA sequence, the synthesis of the gene, the cloning procedure up to the purified protein and even polyclonal antibodies targeted against the protein.

gene cloning expression purification immunisation antibody

Expert support will be provided throughout the project, starting with design and de novo synthesis of the gene using an optimized codon usage for the expression system chosen. In addition, Medigenomix is able to isolate and clone genes from any tissue and from a variety of organisms.

Using Medigenomix' well established mutagenesis technology, a sequence can be easily adapted according to the project outline. In any case will the DNA construct be verified via DNA sequencing. Protein expression can be performed in E. coli, B. subtilis or other expression systems. After a successful pilot expression, the preparative scale will be performed. The latter is followed by protein purification and characterisation according to projects needs. Proteomics expert Toplab offers a variety of methods like multi-dimensional chromatography, free-flow electrophoresis, peptide mass fingerprint (MALDI-MS) and protein sequencing.

Even more detailed characterisations of the protein are available, e.g. identification of post-translational modifications. The service is completed by an immunisation service that leads to polyclonal antibodies directed against the purified protein.

All steps are documented in a detailed quality report.

The cooperation of the two well established biotech service providers allows use of a broad spectrum of molecular biological as well as protein analytical techniques to fulfil all customer's needs.

Medigenomix Acquires an Exclusive License for the Use of further Genetic Polymorphisms in G-Proteins for Applications in Pharmacogenetics

Medigenomix, a subsidiary of the Eurofins Group, recently signed an exclusive license agreement with the University of Duisburg-Essen for the use of Single Nucleotide Polymorphisms (SNPs) in the human GNAS Gene to predict the risks for diseases, the natural course of common disorders and the response to therapies. The cooperation and license negotiations were lead by PROvendis GmbH, the agency for patent utilisation of 24 universities in North Rhine Westphalia.

Medigenomix introduces BLAST extension for the E-SEQUENCE system

The proven online ordering and data transfer system E-SEQUENCE has been expanded by a bioinformatics component. With this new tool our clients have the possibility to get their BLAST alignments in addition to their sequence results – just with a mouse click.

For this purpose E-SEQUENCE has been linked with a standalone BLAST system as known from the National Centre for Biotechnology Information, NCBI. The clear advantage is that our clients do not have to send their sequences via the insecure internet or use special software to get their BLAST alignments for the analysed sequences.

Via secure socket layer the user is able to download the sequence results together with the BLAST alignments in html format. That means the analysed sequence data is absolutely secure, no third party’s access is possible.

The new system allows a fast handling of BLAST alignments in a most secure and comfortable way and guarantees highest efficiency with the dealing of your sequence data.
