Medigenomix offers a reduction of up to 50% of the operating time for backcrossing programs by genotyping with microsatellite markers. By application of approx. 90 markers, which are evenly distributed over the genome, the mouse lines C57BL/6, C3H, 129/Sv, BALB/c and DBA/2 can be differentiated from each other. In every generation the best suited individuals for mating are selected by genotyping thus accelerating the whole process significantly. By this direct selection of the breed animals you can receive mice, which have the genotype of the desired mouse line, after six generations.
Mapping of Mutations, Linkage Analysis
A further area of application of the microsatellite analysis is the mapping of mutations. By two to threefold marker assisted backcrossing of mice, which are descended from mutated founder individuals, on another line (e.g. C3H ENU treated mice on C57BL/6) the chromosome, on which the mutation is situated, can be identified. By using more specific markers in the next steps, which are situated on the same chromosome, a finemapping is executed. In the best case, this limits the localization of the gene locus concerned on 0,1 cM. Finally candidate genes of this region can be cloned and sequenced directly.
